Restriction Isotyping for the detection of Apolipoprotein E isomers.

Objective: To develop a simple, affordable and sensitive method for the detection of Apolipoprotein E isoforms and to identify its population frequency in our region (Jordan). Design : A retrospective study. Place and Duration: The study was conducted as a part of training program held in UK during the period 1995-96. Subjects and Methods: Twenty-five venous blood samples were obtained from normal individuals into EDTA containing tubes. Genomic DNA was isolated from human leukocytes according to the Poncz method. Genomic DNA was amplified with specific primers that included the polymorphic region of amino acids 112 and 158. The amplification products were digested with the restriction enzyme Cfo 1. The digested products were separated on Nusieve agarose - agarose gel electrophoresis, then the gel was stained with ethdium bromide for visualizing the different genotypes. Results: Because of the potential neurotoxicity to the technician and difficulty of handling of polyacrylamide gel electrophoresis, our described method of apo E genotyping on Nusieve agarose-agarose is affordable, suitable for screening program, high resolution of bands, easy to prepare and handle and more favorable for routine analysis as also not time-consuming.
