Abnormalities in the p53 pathway are common in human malignancies In view of the known roles of p53 in mediating growth arrest and apoptosis the mutational state of the p53 gene should be a useful prognostic indicator and should relate to response to radiotherapy or conventional genotoxic chemotherapies. In the future. p53 mutation may provide a novel strategy for therapeutic intervention. However, despite numerous studies, p53 has not proved useful in a clinical setting. We review some recent advances that improve the ability of pathologists to delineate abnormal p53 function in routine pathological material. These techniques should improve the usefulness of assessing p53 in clinical samples and may help to direct clinical decisions based on conventional or novel therapies.
