Nephrotoxicity of Tobramycin alone and in combination with Cefotaxime in a group of Rabbit.

Serious toxicity is a major limitation to usefulness of aminoglycosides. The most important targeting organ is kidney, inner ear and neuromuscular junction. Cephalosporin has been implicated as potentially nephrotoxic agent, although they are not nearly as toxic as to the kidney as are amino glycosides. Aim: The aim of this study was to compare the nephrotoxic effect of tobramycin alone and in combination cephotaxime on rabbit renal function. Materials and Methods: 24 male rabbits were included in the study. Rabbits were divided into three groups. first group received saline, the 2nd group tobramycin and to 3rd tobramycin in combination with cephotaxime Blood samples were drawn on 10th, 16th and 22nd day of infusion of drug. Level of serum creatinine was estimated Increased level of serum creatinine was observed on all days (i.e. of 10th, 16th, and 22nd day) but maximum rise was seen on 16th, day of blood collection. Nephrotoxicity was confirmed by histological study of cortex of kidney. It was observed that renal dysfunction was more in case of combined drug therapy as compared to tobramycin alone. Conclusion: Combination doses of antibiotics (aminoglycosides and cephalosporin) has the potential for systemic toxicity.
