Significance of HER-2/neu Oncoprotein Overexpression on node positive invasive Breast Cancer.

Objective: To determine the frequency of HER-2/neu oncoprotein overexpression in lymph node-positive invasive breast carcinoma and correlate it with various standard prognostic parameters. Design: A cross-sectional study. Place and Duration of Study: Department of Pathology, Basic Medical Sciences Institute, Jinnah Postgraduate Medical Center, Karachi, Pakistan. Subjects and Methods: Sixty-one formalin fixed paraffin embedded blocks of diagnosed node-positive invasive breast carcinoma were retrieved and 5mm thick sections were cut and stained with H&E for the review of diagnosis and grading. The immunohistochemical staining was done on 4mm thick sections by using Polyclonal Rabbit anti HER-2 ZYMED USA, and ZYMED 2nd generation LAB-SA immunodetection system to see the HER-/neu overexpression. Results: Section containing > 50% of tumor cells exhibiting intense circumferential cell membrane staining scored as positive. 33% positive overexpression was observed. A statistically significant correlation (P<.05) was found only between HER-2/neu overexpression and size of tumor. Conclusion: Positive cases can get benefit from Herceptin therapy, especially non-responders to other drugs.
