Evaluation of the efficacy of Xylose-Lysine-Tergitol-4 (XLT4) agar for the isolation of Salmonella species and its comparison with Xylose-Lysine-Deoxycholate (XLD) agar & Salmonella Shigella (SS) agar.

The efficacy of Merck`s XLT4 agar and salmosyst broth has been checked for the isolation of salmonella from 100 red chili samples and 30 Blood Samples, against FDA approved selenite broth, SS agar and XLD agar. Two protocols were followed for that purpose. For XLT4 testing we used the protocol according to Merck`s Manual while for SS agar and XLD agar testing we used FDA`S approved protocol. Final identification of isolates was made by using API-strips. We got 50% Klebsiella Spp., 16 % Salmonella species, 24% Shigella species & 10% Ecoli species on XLT4 agar no any other bacteria grown on XLT4 agar. On SS agar we got 29% Klebsiella species, 12% Salmonella species, 37% Shigella species, 09% E.coli species & remaining 13% were other bacteria. On XLD agar we got 45% Klebsiella species, 06% Salmonella species, 18% Shigella species, 25% E.coli & 06% other bacteria. Out of 30 blood samples isolation rate for salmonella on XLT4 was 55%. We observed increased rate of accompanying bacteria on SS & XLD agar as compare to them XLT4 appeared best for the isolation of Salmonella i.e. 16%, while on XLD it was 06% & on SS agar it was 12% for Salmonella. Also find that Salmonella colonies were really very easy to identify on the basis of their colonial characters & H2S on XLT4 agar.
