Detection of BCR-ABL gene rearrangement by polymerase chain reaction.

Philadelphia (Ph) chromosome is a hallmark of chronic myeloid leukaemia (CML). As a result of the translocation between chromosome 9 and 22 a new fusion gene (BCR-ABL) is created which can be detected by polymerase chain reaction (PCR). In this study ten untreated patients with Ph positive CIVIL were tested by reverse transcription single step PCR (RT-PCR) using a combination of various primers for the BCR and ABL genes. The results showed p210 BCR-ABL gene rearrangements in all of the patients. The PCR based method is sensitive and quick. It can be used for the detection of Ph negative cases with BCR-ABL gene rearrangements. It can also be used in the typical Ph positive patients on treatment for detection of minimal residual disease (MRD).
