Effect Of Alprazolam On Volume Of Distribution And Plasma Concentration In Renal Insufficiency.

The volume of distribution and plasma concentration of alprazolam were evaluated in fifteen patients suffering from renal insufficiency. For this purpose, tab. Alprazolam 0.25 mg, P. O. was given, to five healthy, normal (control) subjects and fifteen renal insufficient patients, twice a day for a period of 21 days. Plasma concentration and Vd of alprazolam were assessed on day seven and twenty one. Among control group, there was no remarkable change noticed in plasma concentration and Vd on day seven and 21. Same was true among renal group on both days. But when renal group was compared with controls on day seven and twenty one, there was a great alteration of Vd with less noticeable change in plasma concentration values. Therefore, it was concluded that long term alprazolam therapy produces significant effect on Vd in renal insufficient patients with less marked effect on plasma concentration when compared with controls.
